sgRNA synthesis

Service introduction


       sgRNA is a high-purity RNA synthesized in vitro that can bind to Cas9 protein to form a complex. The complex can be introduced into cells by electroporation, liposome transfection, or microinjection, and then enters the cell nucleus through the nuclear localization signal peptide (NLS) to perform gene editing.

        Compared with other methods of introducing editing elements, the gene editing method of introducing Cas9/sgRNA complex has several important advantages:


        All elements are in mature form, and do not need to be transcribed and translated. Therefore, there are no problems such as codon incompatibility, transcription regulation, and low transcription efficiency. It is suitable for various cells of various organisms, including stem cells and fertilized eggs, to maximize the success of editing.


        There is no risk of integration of editing elements into the genome. Cas9 and sgRNA are both metabolized by cells and will eventually be metabolized and disappear without leaving a trace. This is especially important in situations where animal trials or clinical trials are required. Similarly, it is also critical if multiple rounds of editing/screening are required.


        The introduction of the Cas9/sgRNA complex has higher editing specificity.
